Missed a Skytrofa Dose? The 2-Day Rule (FDA Label)
[Missed dose · Skytrofa]

I missed a Skytrofa dose. What should I do?

lonapegsomatropin-tcgd (weekly growth hormone) · Once weekly injection, any time of day · Label checked

Short answer: The Skytrofa label says to give a missed dose as soon as possible, but not more than 2 days after the scheduled day. If more than 2 days have passed, skip it and give the next dose on the regular day. The label also allows a dose up to 2 days early, and at least 5 days must pass between doses.

What does the Skytrofa label say about missed doses?

From the FDA label on DailyMed. Numbers in brackets are label section numbers.
SituationWhat the label says
ScheduleOnce weekly, any time of day (2.1, 17)
Up to 2 days after the scheduled dayGive it as soon as possible (2.4)
More than 2 daysSkip it; give the next dose on the usual day (2.4)
Flexible window2 days before or 2 days after the scheduled day (2.4)
Minimum time between dosesAt least 5 days (2.4)

What if I am 2 days late or less?

Section 2.4 of the Skytrofa prescribing information says to give a missed dose as soon as possible and not more than 2 days after the missed dose. Weekly dosing then resumes on the usual day.

Example: your Skytrofa day is Thursday and you remember on Saturday. That is 2 days, so under the label you would inject Saturday and the next one on Thursday. Saturday to Thursday is 5 days, which meets the 5-day minimum.

What if more than 2 days have passed?

Section 2.4 says to skip the dose and give the next one on the regularly scheduled day. If you remember on Sunday (3 days late), you would skip it and inject on Thursday.

Can I take Skytrofa early?

Yes. Section 2.4 says that to avoid missed doses, Skytrofa can be taken 2 days before or 2 days after the scheduled day. This is the only label in this set that names an early window. If you change your regular day, the label says at least 5 days must pass between the last dose and the new day.

How long does a Skytrofa dose last?

Skytrofa releases somatropin slowly. The label gives a mean half-life of about 30.7 hours for lonapegsomatropin in children, and about 25 hours for the somatropin it releases (section 12.3).
