Short answer: The progesterone injection label gives 5 to 10 mg into a muscle for six to eight consecutive days for amenorrhea, with withdrawal bleeding expected 48 to 72 hours after the last injection. For functional uterine bleeding it gives 5 to 10 mg daily for six doses. Each mL holds 50 mg. The label has no missed-dose rule and gives no dosing for any other use.
This is what the FDA label says. Follow your prescriber. Your prescriber may use a different dose or pace than the label's standard schedule.
What Progesterone Injection is approved for: Progesterone injection is approved for amenorrhea (periods that have stopped) and abnormal uterine bleeding caused by hormonal imbalance, when there is no physical cause such as fibroids or uterine cancer (Indications and Usage). Progesterone injection is given by intramuscular injection only. The label notes it is irritating at the place of injection. A healthcare provider or caregiver gives it, following the prescriber's instructions on dose and method (Dosage and Administration and Patient Information).
Each mL holds 50 mg of progesterone with 10% benzyl alcohol as a preservative in sesame oil, in a 10 mL multiple-dose vial (500 mg per vial). It is checked for particles and color before use (Description and How Supplied).
The table below is the label's schedule, from the Dosage and Administration section. Numbers are copied exactly; the section is in brackets.
| Use | Dose | How long | What the label says to expect |
|---|---|---|---|
| Amenorrhea | 5 to 10 mg | Six to eight consecutive days | Withdrawal bleeding 48 to 72 hours after the last injection if the uterine lining is ready; may be followed by spontaneous normal cycles |
| Functional uterine bleeding | 5 to 10 mg daily | Six doses | Bleeding may be expected to stop within six days |
For functional uterine bleeding with estrogen, progesterone starts after two weeks of estrogen therapy. If menstrual flow starts during the course of progesterone injections, they are stopped.
The exam before treatment includes the breasts and pelvic organs and a Pap smear.
The vial is stored at 68 F to 77 F (20 C to 25 C) and not used after the expiration date on the label.
The progesterone injection label gives no missed-dose rule. Neither the prescribing information nor the Patient Information says what to do if a dose is missed; the Patient Information says to follow your doctor's instructions on dose and how it is injected.
In Dose Track you set up Progesterone Injection as a protocol with the dose and schedule your prescriber gave you. A protocol can hold dose steps, each with its own dose and length in days or weeks, so a step-up plan like the one on the label can be entered once and the app moves to the next step on its own. Reminders follow the schedule, and every dose is logged with the date and time, so you can see which step you are on and when your last dose was. The app follows what you enter; it does not choose or change a dose.
