Short answer: The Addyi label dose is one 100 mg tablet by mouth once daily at bedtime, because daytime dosing raises the risk of low blood pressure, fainting and sleepiness. After one or two drinks, the dose waits at least two hours; after three or more, it is skipped. Addyi is stopped after 8 weeks if HSDD has not improved. A missed dose is skipped until the next bedtime.
This is what the FDA label says. Follow your prescriber. Your prescriber may use a different dose or pace than the label's standard schedule.
What Addyi is approved for: Addyi is approved for women under 65 with acquired, generalized hypoactive sexual desire disorder (HSDD): low sexual desire that causes marked distress or relationship difficulty, began after a time without such problems, and is not due to another medical or mental health condition, relationship problems, or a medicine or drug (section 1). It is not for men and not for enhancing sexual performance.

Addyi is a 100 mg tablet taken by mouth once a day at bedtime (sections 2.1 and 3).
It is taken only at bedtime because taking it during waking hours raises the risks of low blood pressure, fainting, accidental injury and sleepiness (section 2.1).
After one or two standard drinks, the bedtime dose waits at least two hours; after three or more drinks that evening, the dose is skipped; and no alcohol is drunk after the dose until the next day (boxed warning, section 5.1).
The table below is the label's schedule, from the Dosage and Administration section. Numbers are copied exactly; the section is in brackets.
| Situation | What the label says |
|---|---|
| Dose | 100 mg orally once per day at bedtime |
| One or two standard drinks that evening | Wait at least two hours after drinking before taking Addyi at bedtime |
| Three or more standard drinks that evening | Skip the Addyi dose that evening |
| After the bedtime dose | No alcohol until the following day |
| Missed bedtime dose | Take the next dose at bedtime on the next day; do not double the next dose |
| No improvement | Discontinue after 8 weeks if HSDD symptoms have not improved |
| Coming off a moderate or strong CYP3A4 inhibitor | Start Addyi 2 weeks after the last dose of the inhibitor |
| Starting a moderate or strong CYP3A4 inhibitor | Start the inhibitor 2 days after the last dose of Addyi |
Activities that need full alertness, such as driving or operating machinery, are avoided until at least 6 hours after a dose and until the person knows how Addyi affects them.
If a moderate or strong CYP3A4 inhibitor must be started within 2 days of stopping Addyi, the person is watched for low blood pressure and fainting.
CYP3A4 inducers such as rifampin, carbamazepine, phenytoin and St. John's Wort sharply lower flibanserin levels, and using them with Addyi is not recommended. Digoxin levels are monitored more closely.
Addyi is contraindicated with any degree of liver impairment.
In people who are CYP2C19 poor metabolizers, side effects such as low blood pressure are monitored more closely.
Addyi is not recommended while breastfeeding.
If a bedtime dose is missed, the next dose is taken at bedtime the next day, and the next dose is not doubled. The Medication Guide adds: skip the missed dose, and do not take Addyi the next morning.
More on this: missed a dose of Addyi.
After a moderate or strong CYP3A4 inhibitor, Addyi is started 2 weeks after the inhibitor's last dose. Before starting such an inhibitor, Addyi is stopped at least 2 days earlier.
In Dose Track you set up Addyi as a protocol with the dose and schedule your prescriber gave you. A protocol can hold dose steps, each with its own dose and length in days or weeks, so a step-up plan like the one on the label can be entered once and the app moves to the next step on its own. Reminders follow the schedule, and every dose is logged with the date and time, so you can see which step you are on and when your last dose was. The app follows what you enter; it does not choose or change a dose.
