Short answer: Orforglipron has a half-life of about 29 to 49 hours. Levels peak 4 to 8 hours after a dose. People vary, so treat these as estimates.
Orforglipron is a GLP-1 receptor agonist taken as a once-daily tablet. It was FDA approved in 2026 as Foundayo for weight loss and keeping weight off in adults with obesity, or with overweight and at least one weight-related condition.
Also known as: Foundayo.
Foundayo comes as 0.8 mg, 2.5 mg, 5.5 mg, 9 mg, 14.5 mg and 17.2 mg tablets.
The FDA Foundayo label doses it once a day by mouth, with or without food. It starts at 0.8 mg, goes to 2.5 mg after at least 30 days, then to 5.5 mg after at least 30 more days. After that it may be raised to 9 mg, 14.5 mg or 17.2 mg, each step at least 30 days apart, based on response and side effects; the maximum is 17.2 mg (9 mg with a strong CYP3A4 inhibitor). After 7 or more missed doses in a row, the label says to restart at a lower dose. It should not be combined with another GLP-1 drug.
The label carries a boxed warning about thyroid C-cell tumors and is contraindicated with a personal or family history of medullary thyroid cancer or MEN 2. People on oral birth control are advised to switch to a non-oral method or add a barrier method for 30 days after starting and after each dose increase. In two 72-week trials, mean weight change was −7.4%, −8.3% and −11.1% on 5.5, 9 and 17.2 mg versus −2.1% on placebo in 3,127 adults without diabetes, and −5.1%, −7.0% and −9.6% versus −2.5% in 1,613 adults with type 2 diabetes. Severe stomach and gut side effects were reported in about 3% on orforglipron versus 1% on placebo.
The FDA Foundayo label gives an elimination half-life of about 29 to 49 hours and peak levels 4 to 8 hours after a dose, with steady state after about 1 week of daily dosing. Bioavailability was 77% at 0.8 mg, and food had no clinically relevant effect.
