Short answer: Avanafil has a half-life of about 5 hours. Levels peak 30 to 45 minutes (fasted) after a dose. People vary, so treat these as estimates.
Avanafil is a PDE5 inhibitor, the same drug class as sildenafil and tadalafil. It helps blood flow into the penis during sexual stimulation. It is sold as Stendra (Metuchen Pharmaceuticals), a tablet taken as needed for erectile dysfunction.
Also known as: Stendra.
Stendra comes as 50 mg, 100 mg and 200 mg tablets, taken with or without food.
The Stendra label approves avanafil for erectile dysfunction in adult men. Label dosing starts at 100 mg as needed, as early as about 15 minutes before sex; the dose can go up to 200 mg (also about 15 minutes before) or down to 50 mg (about 30 minutes before), at the lowest dose that works, no more than once a day. With an alpha-blocker it starts at 50 mg. Sexual stimulation is needed for it to work.
The label has no boxed warning. It is not used with any nitrate or with riociguat or vericiguat, because blood pressure can drop sharply; if a nitrate is needed in an emergency, at least 12 hours should pass after the last dose. It is not recommended within 6 months of a heart attack, stroke or serious arrhythmia, or with strong CYP3A4 inhibitors. Other warnings cover erections over 4 hours, sudden vision or hearing loss, and alcohol. Headache was 10.5% on 200 mg vs 1.7% on placebo. Full numbers are on the Stendra side effects and Stendra dosing schedule pages. Comparisons: Stendra vs Viagra and Stendra vs Cialis. Storage: ED tablets.
The Stendra label says avanafil is absorbed quickly, with a median time to peak of 30 to 45 minutes fasted. A high-fat meal delayed the peak by about 1.1 to 1.25 hours and lowered it by 39%. The terminal half-life is about 5 hours.
