PE-22-28 is a synthetic seven-amino-acid peptide made by French researchers as a shorter, longer-lasting version of spadin, a natural peptide that blocks the TREK-1 potassium channel in the brain. It has been studied only in cells and mice and is not approved for human use.
Also known as: PE 22-28, Spadin analog.
We could not confirm a standard vial size for PE-22-28, so check the label on yours. The 10 mg vial in the example below is only there to show the math.
Here is the math for a 10 mg vial mixed with 2 mL of bacteriostatic water, drawing 500 mcg with a U-100 insulin syringe (1 mL = 100 units).
10 mg ÷ 2 mL = 5 mg/mL500 mcg ÷ 5 mg/mL = 0.1 mL0.1 mL × 100 = 10 units| Amount | Volume | U-100 syringe |
|---|---|---|
| 250 mcg | 0.05 mL | 5 units |
| 500 mcg | 0.1 mL | 10 units |
| 1 mg | 0.2 mL | 20 units |
Adding more water doesn't change how much PE-22-28 is in the vial. It only spreads it over more liquid, so each draw is bigger and easier to read.
| Water added | Concentration | Volume | U-100 syringe |
|---|---|---|---|
| 1 mL | 10 mg/mL | 0.05 mL | 5 units |
| 2 mL | 5 mg/mL | 0.1 mL | 10 units |
| 3 mL | 3.33 mg/mL | 0.15 mL | 15 units |
| 5 mL | 2 mg/mL | 0.25 mL | 25 units |

Nothing yet: we could not find any published study or registered trial that gave PE-22-28 to people. Its parent peptide, spadin, had antidepressant-like effects in mice (Mazella 2010). In the paper that introduced PE-22-28 (Djillani 2017), it blocked TREK-1 far more strongly than spadin in cells, had antidepressant-like effects in mouse behavior tests after 4 days, increased new nerve cells, and its effect lasted up to 23 hours in mice versus 7 hours for spadin. Mouse results often do not carry over to people.
We could not find a published human half-life for this compound. Figures quoted online usually come from animal or cell studies, so we do not list one here.
