Short answer: Dasatinib has a half-life of about 3 to 5 hours. Levels peak 0.5 to 6 hours after a dose. People vary, so treat these as estimates.
Dasatinib is a kinase inhibitor, a targeted cancer drug sold as Sprycel tablets for two types of leukemia (Philadelphia chromosome-positive CML and ALL). Combined with quercetin ("D+Q"), it has been studied off-label as a senolytic, a drug meant to clear senescent ("zombie") cells that build up with age.
Also known as: Sprycel.
Sprycel comes as 20 mg, 50 mg, 70 mg, 80 mg, 100 mg and 140 mg tablets.
The FDA Sprycel label approves dasatinib for adults with newly diagnosed or treatment-resistant Philadelphia chromosome-positive chronic myeloid leukemia (CML) or resistant acute lymphoblastic leukemia (ALL), and for children 1 year and older with chronic-phase CML or, with chemotherapy, newly diagnosed ALL. The adult starting dose is 100 mg once a day for chronic-phase CML and 140 mg once a day for advanced CML or ALL. Warnings include low blood counts, bleeding, fluid retention (including fluid around the lungs), heart problems, pulmonary arterial hypertension, QT prolongation, severe skin reactions, tumor lysis syndrome, harm to an unborn baby and liver injury. Stomach-acid reducers (H2 blockers and proton pump inhibitors) lower its absorption, and grapefruit juice and strong CYP3A4 blockers raise its levels.
Senolytic use is off-label and the trials are small. In an open-label pilot in 14 people with idiopathic pulmonary fibrosis (Justice, 2019), dasatinib 100 mg plus quercetin 1,250 mg a day, 3 days a week for 3 weeks, was followed by better walking and chair-stand scores. In 9 people with diabetic kidney disease (Hickson, 2019), 3 days of dasatinib 100 mg plus quercetin 1,000 mg lowered markers of senescent cells in fat and skin 11 days later. A placebo-controlled pilot in 12 people with pulmonary fibrosis (Nambiar, 2023) found D+Q feasible and generally tolerated, with more mild side effects than placebo (sleep problems and anxiety in 4 of 6 versus 0 of 6) and no clear difference in function. These are study protocols, not personalized advice; confirm any dosing with your prescriber.
The FDA Sprycel label gives a mean terminal half-life of 3 to 5 hours, with peak levels 0.5 to 6 hours after a dose. A dose of the proton pump inhibitor omeprazole taken 22 hours earlier cut dasatinib exposure by 43%. The 2019 kidney study called its few-day course "hit-and-run" treatment, noting that both drugs have half-lives under 11 hours.
